Film, kötü ve tatsız şeylerden hep korunmuş, sıradan bir evcil hayvan hayatı yaşayan bukalemun Rango'nun, büyük bir kimlik bunalımı yaşarken başından geçen komik dönüşüm hikayesini konu ediyor. Sonuçta hayattaki tek amacınız kalabalığa uyum sağlamaksa hedefiniz ne kadar yüksekte olabilir ki? Rango kazara kendini gözüpek, eşkıyaların ve kanunsuzların olduğu, nüfusunu çölün en sahtekar ve üçkâğıtçı yaratıklarının oluşturduğu Toprak kasabasında bulunca pek de cesur olmayan bu kertenkele, dikkat çektiğini fark eder. Rango, eskiden numarasını yaparken şimdi kahraman olmaya başlamıştır.
